It depends on the story, location where the level is taking place, atmosphere of the level (game), other enemies...

Well, a lot of things! Tell something about your game and maybe I (or someone else) can think of something

I know only 4 mortal bad guys that are the hardest to kill:
1) Demigod2
2) Demigod3
3) Scorpion
4) Horseman

Of course there might be more than these 4...

Thanks! But what are Demigod2 and 3? I've used the one in Cleopatra's Palaces for normal enemy already, as well as the Horseman.

You don't know these?
If yoy played Cleopatra's palaces, Demigod3 were the two guys who guarded Cleo's throne.
Demigod2 is quite harder to beat (I think), it guarded the star-shaped keys in the pyramid levels in TR:LR
